Potential unreliability of ALK variant allele frequency in the efficacy prediction of targeted therapy in NSCLC

Frontiers of Medicine 2023, Volume 17, Issue 3,   Pages 493-502 doi: 10.1007/s11684-022-0946-x

Abstract: Anaplastic lymphoma kinase (<i>ALKi>) is the most common fusion gene involved in non-small cell lungIn this work, we investigated whether the variant allele frequencies (VAFs) of <i>ALKi> fusions canALKi> positive.ALKi> subclonality with crizotinib efficacy was assessed. Results suggest that the <i>ALKi> VAF determined by hybrid capture-based NGS is probably unreliable

Frontiers of Agricultural Science and Engineering 2017, Volume 4, Issue 1,   Pages 48-58 doi: 10.15302/J-FASE-2016114

Abstract: Wheat stripe rust caused by f. sp. is one of the most devastating diseases of wheat worldwide and resistant cultivars are vital for its management. Therefore, investigating the heterozygosity of the pathogen is important because of rapid virulence changes in isolates heterozygous for avirulence/virulence. An isolate of f. sp. was selfed on to determine the heterozygosity for avirulence/virulence loci. One hundred and twenty progeny isolates obtained from this selfing were phenotyped using 25 lines of wheat containing genes and genotyped with 96 simple sequencing repeat markers, with 51 pathotypes and 55 multi-locus genotypes being identified. All of these were avirulent on lines with , , , and and virulent on lines with , and , indicating that the parental isolate was homozygously avirulent or homozygously virulent for these loci. Segregation was found for wheat lines with , , , , , , , , , , , , , , , and . The 17 cultivars to which the was identified as heterozygous with respect to virulence/avirulence should not be given priority in breeding programs to obtain new resistant cultivars.

Detection and monitoring prostate specific antigen using nanotechnology approaches to biosensing

Frontiers of Chemical Science and Engineering 2020, Volume 14, Issue 1,   Pages 4-18 doi: 10.1007/s11705-019-1846-8

Abstract: Prostate cancer has a high incidence in men and remains the second cause of mortality due to cancer. As the development of the disease is greatly correlated to age, the identification of novel detection methods reliable, efficient, and cost effective is a matter of significant importance in the ageing population of western societies. The detection of the prostate specific antigen (PSA) in blood samples has been the preferred method for the detection and monitoring of prostate cancer over the past decades. Despite the indications against its use in massive population screening, PSA still remains the best studied biomarker for prostate cancer and the detection of its different forms and incorporation in multiplexed designs with other biomarkers still remains a highly valuable indicator in the theranostics of prostate cancer. The latest developments in the use of nanomaterials towards the construction of PSA biosensors are reviewed hereby. The incorporation of gold nanoparticles, silica nanoparticles and graphene nanostructures to biosensing devices has represented a big leap forward in terms of sensitivity, stability and miniaturization. Both electrochemical and optical detection methods will be reviewed herein for the detection of PSA.

Development of Perennial Wheat Through Hybridization Between Wheat and Wheatgrasses: A Review Review

Engineering 2018, Volume 4, Issue 4,   Pages 507-513 doi: 10.1016/j.eng.2018.07.003

Abstract:

Wheatgrasses (Thinopyrum spp.), which are relatives of wheat (Triticum aestivum L.), have a perennial growth habit and offer resistance to a diversity of biotic and abiotic stresses, making them useful in wheat improvement. Many of these desirable traits from Thinopyrum spp. have been used to develop wheat cultivars by introgression breeding. The perennial growth habit of wheatgrasses inherits as a complex quantitative trait that is controlled by many unknown genes. Previous studies have indicated that Thinopyrum spp. are able to hybridize with wheat and produce viable/stable amphiploids or partial amphiploids. Meanwhile, efforts have been made to develop perennial wheat by domestication of Thinopyrum spp. The most promising perennial wheat–Thinopyrum lines can be used as grain and/or forage crops, which combine the desirable traits of both parents. The wheat–Thinopyrum lines can adapt to diverse agricultural systems. This paper summarizes the development of perennial wheat based on Thinopyrum, and the genetic aspects, breeding methods, and perspectives of wheat–Thinopyrum hybrids.
